I am planning a trip to vegas in february and would like to stay 2 nights (check in thurs 2/21- check out sat 2/23) but I'm also possibly considering staying an additional 2 nights (check in sat 2/23-check out mon 2/25). it is my understanding that i can combine nights but, would i have to buy 2 of the "option 2" groupons or 1 "option 2" and 1of either "option 2 or 1"?

Each voucher counts for the first day it is being used, so if you're checking in Thursday, you need an option 2 for Thursday. If you stay longer, through Tuesday, you'll need to get a option 1 Sunday - Thursday Groupon.
